Zoloft Birth Defects: Epispadias

The prescription antidepressant Zoloft has been linked to a number of birth defects, including epispadias.

An epispadias is a rare type of malformation of the penis in which the urethra does not extend completely through the penis and ends in an opening on the upper aspect. The birth defect can also develop in females when the urethra is place too far anteriorly. The defect affects approximately 1 in 120,000 male and 1 in 500,000 female births.

Epispadias is an uncommon and partial form of a group of failures of abdominal and pelvic fusion in the first months of embryogenesis known as the exstrophy – epsipadias complex. While epispadias is present in all cases of exstrophy, it can also occur in isolation as the least severe form of the complex spectrum.

Most cases of epispadias involve a small and bifid (double) penis, which requires surgical closure soon after birth, commonly including a reconstruction of the urethra. When Epispadias is part of a larger exstrophy, not only the urethra but also the bladder or perineum may be open and exposed, requiring closure.
